You can set the time interval until you will next be asked for your master password in the Security tab of the 1Password preferences, as shown in this screen capture.
I don't think there is a way to keep from having to enter it when you restart your system, though.

in 3.0 ver. I rarely had to enter Master Pass on start up. Why is it not available in ver 4.0. Help

In 1PW 3 the master password could be stored in the user keychain. This was removed in 1PW 4, presumably because it was too great a security risk. Entering the password once at startup is not much of an effort and improves security.

Like the hawkmoth and danco mentioned, you will always need to enter your master password after a restart. This is intended behaviour in 1Password 4.
1Password 3.8 had a special option that allowed you to store the master password in the OS X keychain. We removed this option in 1Password 3.9 (Mac App Store version) for several reasons. The most important one was that people who store the master password in the OS X keychain tend to forget it because they are never forced to type it. Forgetting the master password is the same as losing all 1Password data. In addition, there are also security concerns with storing your master password anywhere, such as the OS X keychain.

I moved your post here since it's related more to this discussion about keeping 1Password 4 unlocked.
Which version of 1Password are you currently using? If it's from the Mac App Store, have you ever ran a Agile website/beta version? One possible reason for the behavior you've reported is that the 1Password 4 helper processes from the Mac App Store and website (or beta) versions are both running. To check that, please run this command in a Terminal window and post its output:
launchctl list | grep agile
I have unchecked all security preferences on 1P4 Mac Client. But still the browser plugin locks.
Can you mention a specific example of when you're unexpectedly discovering 1Password locked after you've unlocked it?

One question - Is this browser plugin locking control by mac client and mac mini ?
Locking is handled by 1Password mini, which powers the interface for both the 1Password browser extensions and main application.
1Password 4 is either fully locked or unlocked, adjustable with the Auto-lock options under the Security tab of Preferences. There are no separate locking options for the main application, 1Password mini, and browser extensions.
